1) Advantage or edge. In lowball, it might be said of a conservative player, "When you're both drawing, he's usually got the best of it." For this sense, the opposite of worst of it.

2) An edge gained by cheating; often preceded by taking or take. "He's never in a game unless he's taking the best of it."
